Usyk is scheduled to confront Spong in Chicagos Wintrust Arena in his first departure when he defended all of four world cruiserweight titles.
Spong, 34, competed in kickboxing before winning his first 14 fights as a boxer and spent a lot of his combat career in mixed martial arts.
Vada requires promoters or fighters to pay for testing for the length of a struggle build-up and operates in boxing and MMA. It said Spong was a part of its testing programme on 20 September.
